Using an opioid drug(1) to induce(2) hibernation(3) in rats(4) reduces the damage caused by an artificial stroke(5), reports a study published today in the open access(6) journal BMC Biology(7). Researchers(8) found that those animals put into a chemical slumber(9) -- a hibernation-like(10) state that cooled their brains(11) -- suffered less behavioral(12) impairment(13) after a period of cerebral artery(14) blockage(15) than control(16) rats.
Cesar Borlongan, PhD(17), a neuroscientist(18) at the University of South Florida Center for Aging(19) and Brain Repair(20) in Tampa, FL, worked with a team of researchers from the National Institutes of Health(21), to investigate the role of the opioid system(22) in brain injury(23) and protection.
“Studies in hibernating(24) and active squirrels(25) have shown that ‘natural hibernation’ has anti-ischemic effects(26) – protecting against the formation of blood clots(27). We’ve shown that a drug that induces hibernation can achieve similar results in the brain,” Borlongan said. “Even a small decrease in the brain’s temperature appears to be neuroprotective(28).”
Borlongan and his colleagues(29) dosed(30) the rats intravenously(31) with [D-ala2,D-leU5]enkephalin (DADLE) (32), a drug from the same pharmaceutical(33) family as morphine(34) and heroin(35). DADLE is used to cryogenically preserve(36) donated(37) organs(38) to keep them viable(39) for transplantation(40). The researchers found that, after an experimental(41) stroke, the pre-treated(42) animals performed better than control rats in a series of(43) behavioral tests. DADLE significantly(44) reduced the size of the stroke, and prevented cell death(45) processes and behavioral abnormalities(46).
“The observation that this substance, previously shown to induce hibernation, improves recovery from cerebral ischemia(47) means it could provide a new pharmacological treatment for stroke,” Borlongan said.
