急性骨髄性白血病細胞
ヒトの癌細胞について初めて全ゲノムが解析された。これまで,癌の遺伝的な原因についての研究はほとんど関連がありそうな遺伝子に焦点が絞られていた。しかし,今回の解析では,1人の急性骨髄性白血病患者の正常な細胞と癌細胞の全DNAを比較した。30億もの塩基対を分析した結果,遺伝子の変異が10個みつかり,そのうちの8個は白血病との関連が知られていない新たな変異だった。これらの変異は癌の増殖に関わる遺伝子や,細胞への薬の送達に関わる遺伝子に起こっていたそうだ。今後,乳癌や肺癌のDNAも同様に分析される予定だという。(吉田素子)
For the first time, scientists have decoded(1) the complete DNA of a cancer(2) patient and traced her disease - acute myelogenous leukemia(3) - to its genetic(4) roots. A large research team at the Genome Sequencing(5) Center and the Siteman Cancer Center at Washington University School of Medicine in St. Louis and Barnes-Jewish Hospital sequenced(6) the genome(7) of the patient - a woman in her 50s who ultimately(8) died of her disease - and the genome of her leukemia cells(9), to identify genetic changes unique to(10) her cancer.
The study is reported in the Nov. 6 issue of the journal Nature.
The pioneering(11) work sets the stage for(12) using a more comprehensive(13), genome-wide(14) approach to unravel(15) the genetic basis of cancer. "Our work demonstrates the power of sequencing entire genomes to discover novel cancer-related(16) mutations(17)," says senior author(18) Richard K. Wilson, Ph.D.(19), director of Washington University's Genome Sequencing Center. "A genome-wide understanding of cancer, which is now possible with faster, less expensive DNA sequencing technology, is the foundation for developing more effective ways to diagnose(20) and treat(21) cancer."
The researchers(22) discovered just 10 genetic mutations in the patient's tumor(23) DNA that appeared to be relevant to(24) her disease; eight of the mutations were rare and occurred in genes(25) that had never been linked to AML(26). They also showed that virtually(27) every cell(28) in the tumor sample had nine of the mutations, and that the single genetic alteration(29) that occurred less frequently(30) was likely the last to be acquired(31). The scientists suspect that all the mutations were important to the patient's cancer.
